Kisspeptin is a neuropeptide encoded by the KISS1 gene and recognized as a pivotal regulator of the hypothalamic-pituitary-gonadal (HPG) axis. Acting on its receptor KISS1R (GPR54), kisspeptin stimulates the secretion of gonadotropin-releasing hormone (GnRH), which in turn modulates downstream reproductive hormone cascades including LH and FSH release. Loss-of-function mutations in KISS1R were among the first identified causes of idiopathic hypogonadotropic hypogonadism in humans.
